What affects our standing with God is that we have accepted Jesus Christ, His Son, as our savior.
After Jesus' death and resurrection we are not told to keep the Sabbath. We are told not to fail to meet with other brothers and sisters in the Lord. We can meet any time, any place, any day of the week. If you esteem one day above another for meeting, that may be o.k for you, but it is not a rule for everyone. Hebrews 10:25/Romans 14.

There again - what is the sabbath? It is a matter of the heart and spirit, not a matter of following a rule (if in fact it is one).
Did you know the Americans are the most benevolent in the world? Check out the countries who came to the rescue after the disasters in Indonesia (88% Muslim), Japan, Filipins, etc. Or better yet, search out the countries that did not help.

According to the letter of the Law, yes, the Pharisees were absolutely correct (if you go back and read the mandates of the Sabbath that Moses laid out). The scripture that you gave doesn't support that but what it does support is that the Pharisees were hypocrites who required the complete following of the Law but didn't do it themselves. That is part of the beauty of Christ approach with them. A lot of people don't realize that Jesus made some of the law more strict than it ever has been because he made it not just a matter of doing and following but a matter of the heart but I digress. The work that wasn't to be done wasn't earning of the daily bread (again, the man who was stoned was not earning any wage for what he was doing) but it was physical labor period. The food that was prepared didn't need to be gathered. So again, by the LETTER of the law (as given by Moses), they shouldn't have been out walking doing anything, let alone picking grain to eat.

I think if you read Titus 3 and James 1:21-27 you might agree that the "works" spoken of is a lifestyle. In our daily living we need to express the nature of our Lord. That we can't accept the Lord as our Savior and then go about our lives living in sin purposely.
There are no rules or methods by which we live righteously. We live righteously in Him, by exercising our spirit.

The boss gives us a day off, a specific day in the week so he may party with the employees.
Some of my colleagues said thankyou very much but I'd rather work because a day off is such a burden. Beside, I see him everyday so what's another party.
Some took the day off but moonlight for the competitors instead of going to the party.
Some asked why don't we do a poll
a. you must go to the party to keep the job.
b. going to the party has any bearing to job security and relationship with the boss.
c. I can't answer it, it's too complicated.
For me and my family, we are going to the party because the boss is like our family - it's too complicated to explain.

If you were to ask does keeping the Sabbath save you then the answer would be an easy no. just as not killing does not save you. we are saved by faith in Gods promise to save through Jesus Christ.
however your question is does keeping the Sabbath "effect" salvation or standing before God. I will answer this with another question.
Does murdering or not murdering or committing adultery etc effect your salvation or standing with God?
The answer is the Same.
